This is only for a graphical login. You do not need to do this to use
su.

Bottom line is you are using the wrong password. Maybe capslock, you
used punctuation instead of digits, or something else.
There is a possibility that you have been hacked and the hacker has
changed the password. Happened to me on a non-root account once.
Finally you may simply have to remove the root password then create a
new one. There are ways to achieve this if you need to.
